Ectonucleotide Pyrophosphatase/Phosphodiesterase 2, also known as Autotaxin, is a secreted lysophospholipase D that hydrolyzes lysophospholipids to produce the signaling molecule lysophosphatidic acid (LPA) in extracellular fluids. Its primary substrate is lysophosphatidylcholine. Enpp2 plays a crucial role in embryonic development, particularly in its expression within yolk sac endoderm cells (VECs). Mice lacking Enpp2 (Enpp2−/−) die at embryonic day 9.5 due to defective yolk sac angiogenesis, exhibiting severe angiogenesis abnormalities and developmental defects, such as impaired phagocytosis and secretion by yolk sac endoderm cells.
