Legumain serves multiple functions in mice, including immune regulation, extracellular matrix remodeling, vitamin D metabolism, and kidney protection. It plays a role in modulating the inflammatory response during renal ischemia-reperfusion injury (IRI) by promoting ferroptosis. Research indicates that mice deficient in Legumain exhibit reduced inflammatory cell infiltration and lower levels of proinflammatory factors like TNF-α and IL-1β in the IRI model, along with significantly less renal tubular damage. Additionally, Legumain is involved in regulating the function of T cells and macrophages. In the tumor microenvironment, for instance, Legumain inhibits osteoblast differentiation by cleaving specific substrates such as TGF-β1 and influences the activity of tumor-associated macrophages (TAMs), thereby impacting tumor progression and metastasis.
