Annexin A5 is a protein with anticoagulant properties that indirectly inhibits the thromboplastin-specific complex, a key component in the blood clotting process. In mice, AnxA5 is involved in a range of complex biological processes, such as bone development, cardiovascular protection, immune system regulation, inflammatory response, and apoptosis. Although mice lacking AnxA5 can survive under normal conditions, their heart and bone structures are susceptible to damage when subjected to repeated mechanical or inflammatory stress. AnxA5 is also crucial for the immune system; its absence leads to a weakened immune response to foreign cells and reduces the ability of macrophages to engulf apoptotic cells. In models of endotoxin shock, AnxA5 enhances survival by inhibiting inflammatory responses mediated by HMGB1.
