Mouse Runt-Related Transcription Factor 2 (Runx2) is a key transcription factor essential for osteoblastic differentiation and skeletal morphogenesis. It acts as a scaffold for nucleic acids and regulatory factors involved in skeletal gene expression. Runx2 is crucial for the regulation of chondrocyte maturation and proliferation, as well as osteoblast differentiation. It directly regulates the expression of Indian hedgehog (Ihh), which enhances chondrocyte proliferation and forms a negative feedback loop with parathyroid hormone-like hormone (Pthlh) through the parathyroid hormone 1 receptor (Pth1r). Runx2 also induces the expression of vascular endothelial growth factor A (Vegfa), secreted phosphoprotein 1 (Spp1), integrin binding sialoprotein (Ibsp), and matrix metallopeptidase 13 (Mmp13) in terminal hypertrophic chondrocytes.
