Rat thyroid stimulating hormone receptor (TSHR) is a 84-kDa G protein-coupled receptor (GPCR) exclusively expressed in rat thyroid follicular cells. It binds thyroid stimulating hormone (TSH), activating cAMP and phosphatidylinositol signaling pathways to regulate thyroid hormone (T3/T4) synthesis, follicular cell proliferation, and iodine uptake. Critical for maintaining systemic metabolic homeostasis, growth, and thermoregulation in rats, it integrates hypothalamic-pituitary-thyroid axis signaling. Dysregulated activation or mutations associate with hyperthyroidism (Graves' disease), hypothyroidism, and thyroid tumors in rat models. As a central mediator of thyroid function and metabolic regulation, rat TSHR is a critical target for studying thyroid pathobiology and developing therapeutics for thyroid disorders.
