Target Information
Rat Iron-Responsive Element Binding Protein 2 (IREB2, also IRP2) is a cytosolic RNA-binding sensor that regulates post-transcriptional iron homeostasis. In low-iron conditions it binds stem-loop IREs within ferritin, ferroportin and transferrin-receptor mRNAs, repressing ferritin/ferroportin translation while stabilizing transferrin-receptor mRNA, thereby increasing cellular iron uptake. When iron is abundant IRP2 undergoes ubiquitination and proteasomal degradation, permitting storage and export gene expression. Expressed ubiquitously, IRP2 controls erythropoiesis, neuro-iron balance and oxidative stress. As the master post-transcriptional regulator of iron metabolism, IRP2 is an important target for studies of anemia, neurodegeneration and metabolic disorders.
