Mouse lipopolysaccharide binding protein (LBP) is a 60-kDa acute-phase glycoprotein predominantly synthesized in mouse hepatocytes and secreted into plasma, with secondary expression in intestinal epithelial cells and macrophages. It binds to lipopolysaccharide (LPS) from Gram-negative bacteria, forming high-affinity complexes that enhance LPS recognition by the CD14-TLR4-MD2 receptor complex on immune cells. This interaction amplifies innate immune responses-promoting pro-inflammatory cytokine secretion, phagocyte activation, and pathogen clearance-while also regulating LPS detoxification to prevent excessive inflammation. Inducible by infection, tissue damage, or inflammatory signals, it acts as a key bridge between bacterial detection and immune activation. As a central mediator of LPS-induced immunity, mouse LBP is a critical target for studying sepsis, inflammatory disorders, and host-pathogen interactions.
