Mouse L-type voltage-gated calcium channel α1S (CACNA1S) is the pore-forming subunit of the dihydropyridine-sensitive skeletal muscle CaV1.1 channel. It conducts calcium influx that couples sarcolemmal depolarization to ryanodine-receptor-mediated sarcoplasmic reticulum calcium release, initiating excitation-contraction coupling. The α1S subunit is expressed exclusively in skeletal muscle and serves as a scaffold for the β, α2δ, and γ accessory subunits. As a critical mediator of muscle contraction and calcium homeostasis, CACNA1S is an important target for studies of muscle physiology, channelopathies, and neuromuscular disorders.
