Mouse KH-domain-containing RNA-binding signal-transduction-associated protein 1 (KHDRBS1/Sam68) is a 55 kDa STAR-family post-transcriptional module whose single KH domain clamps U/A-rich motifs, while N- and C-terminal tyrosine-rich tails dock Src kinases and poly(A) polymerase. A solvent-exposed 18-residue "GSG" linker (aa 140-157) flanks the KH core, presenting a conformational epitope unique among Sam68-like isoforms. Affinity-purified antibodies targeting this linker trace native KHDRBS1 in nuclear speckles, cytoplasmic stress granules, and neuronal dendrites, delivering crisp puncta and single-band specificity on immunoblots with no cross-reaction to SLM-1 or SLM-2.
