Rat 5'-nucleotidase, cytosolic III (NT5C3, cN-III) is a 34 kDa metal-dependent phosphatase whose α-β sandwich active site converts AMP/IMP to adenosine/inosine, balancing nucleotide pools. A solvent-exposed 15-aa "KVE" loop (aa 145-159) between β6 and α4 folds into a β-hairpin unique among rat nucleotidases, presenting a conformational epitope preserved after fixation. Affinity-purified antibodies targeting this loop trace native NT5C3 in hepatic cytosol, renal proximal tubules, and splenic lymphocytes, delivering crisp diffuse staining and single-band clarity on immunoblots with no cross-reaction to NT5C1 or NT5C2.
