Target Information
Rat growth differentiation factor 9 (GDF-9) is a 52 kDa disulfide-linked homodimer whose C-terminal cystine-knot binds BMPR-II and ALK5 on ovarian granulosa cells, driving follicle maturation. A solvent-exposed 15-aa "KVE" loop (aa 102-116) between pre-helix 1 and β2 folds into a β-hairpin unique among rat TGF-β superfamily members, presenting a conformational epitope preserved after activation and fixation. Affinity-purified antibodies targeting this loop trace native GDF-9 in oocyte cytoplasm, antral follicles, and pituitary, delivering crisp punctate granules and single-band clarity on immunoblots with no cross-reaction to BMP-15 or GDF-5.
