Guinea-pig interleukin-4 (IL-4) is an 18 kDa four-helix-bundle cytokine that binds the IL-4Rα/IL-13Rα1 heterodimer on Th2 cells, mast cells and macrophages, triggering STAT6 phosphorylation to drive B-cell class-switching and IgE production. Its solvent-exposed receptor-binding face presents a unique, species-restricted epitope ideal for antibody generation; anti-IL-4 immunoreagents detect both secreted and intracellular forms in lavage, plasma or fixed tissues without cross-reacting with IL-13, offering a specific read-out of Th2-driven inflammation .
