Mouse B-cell CLL/lymphoma 3 (Bcl3) is a 47 kDa ankyrin-repeat protein that threads through DNA-bound NF-κB dimers, stabilizing their nuclear residence and sculpting transcriptional output. A solvent-exposed 20-residue "KEL" loop (aa 280-299) bridging repeats 5 and 6 folds into a β-turn unique among IκB-like regulators, offering a conformational epitope preserved after fixation. Affinity-purified antibodies targeting this loop trace native Bcl3 in germinal-center B cells, splenic follicles, and activated macrophage nuclei, delivering crisp punctate staining and single-band clarity on immunoblots with no cross-reaction to IκBα or IκBε.
