Target Information

Mouse tryptophanyl-tRNA synthetase (WARS1) is a 53 kDa class-I enzyme whose Rossmann fold grips ATP and tryptophan, while an appended KMSKS loop seals the active site during aminoacylation. A solvent-exposed 14-residue "GNE" loop (aa 152-160) within the L-domain exhibits a mouse-specific glycine insertion, presenting a conformational epitope preserved in both cytoplasmic and secreted fragments. Affinity-purified antibodies targeting this loop detect native WARS1 in liver cytosol, brain synaptosomes, and activated macrophages, yielding crisp cytoplasmic puncta and single-band clarity on immunoblots with no cross-reaction to tyrosyl- or leucyl-tRNA synthetase.
