Rat Leucine-Rich Repeat Kinase 2 (LRRK2) is a large, multidomain cytoplasmic kinase highly expressed in brain, lung, and kidney. Its ROC-COR GTPase and kinase modules phosphorylate Rab GTPases and cytoskeletal proteins, regulating vesicle trafficking, autophagy, and microtubule dynamics. LRRK2 activity is enhanced by pathogenic mutations, disrupting neuronal vesicle transport and mitochondrial function. As a central player in cellular trafficking and Parkinsonian pathways, LRRK2 is a key target for studies of neurodegeneration, lysosomal biology, and kinase-related therapeutics.
