Target Information

Protein-glutamine gamma-glutamyltransferase E (TGM3) catalyzes the calcium-dependent formation of isopeptide cross-links between glutamine and lysine residues in various proteins, as well as the attachment of polyamines to proteins. It plays a key role in forming the cornified envelope (CE), a specialized structure consisting of covalently cross-linked proteins beneath the plasma membrane of terminally differentiated keratinocytes. TGM3 is crucial for the development of mouse skin and hair. Research has shown that mice lacking TGM3 exhibit hair abnormalities, such as thinning, twisting, and slower growth, which resemble the characteristics of human uncombed hair syndrome (UHCS).
