Target Information
Mesencephalic astrocyte-derived neurotrophic factor (MANF) selectively supports the survival of dopaminergic neurons in the ventral midbrain and modulates GABAergic transmission to dopaminergic neurons in the substantia nigra. It enhances both spontaneous and evoked GABAergic inhibitory postsynaptic currents in these neurons, while also inhibiting cell proliferation and preventing endoplasmic reticulum (ER) stress-induced cell death. In mice, MANF is highly expressed in the endocrine system, particularly in the pituitary, thyroid, pancreas, adrenal glands, and gonads. Research has shown that MANF deficiency can lead to abnormal development of the anterior pituitary gland in mice, resulting in a reduced number of growth hormone (GH) and prolactin (PRL) secreting cells, and consequently, causing growth retardation and diabetes.
