Target Information
G-protein coupled estrogen receptor 1 (GPER1) is a receptor that binds with high affinity to 17-beta-estradiol (E2), resulting in the rapid and transient activation of various intracellular signaling pathways. It enhances cAMP production, calcium mobilization, and the activation of tyrosine kinase Src, which leads to the release of heparin-bound epidermal growth factor (HB-EGF) and the subsequent transactivation of the epidermal growth factor receptor (EGFR). This activation triggers downstream signaling pathways such as PI3K/Akt and ERK/MAPK. GPER1 is broadly expressed in the mouse brain, particularly in the hippocampus, suggesting its significant role in regulating synaptic plasticity and neuronal activity. In the mouse hippocampus, activating GPER1 can enhance learning and memory by influencing the NR2B/CaMK2/CREB signaling pathway. Conversely, the absence of GPER1 exacerbates MK-801-induced schizophrenia-like cognitive impairment.
