The role of pulmonary surfactant protein A1 (SP-A1) in mice can be examined from several angles. SP-A1 contributes to the pulmonary surfactant system, which is crucial for the innate immunity of the lung. Research indicates that SP-A1 and SP-A2 differ in their ability to enhance macrophage phagocytosis, with SP-A2 being more effective, possibly due to differences in their amino acid sequences. In mice lacking SP-A, infection with Staphylococcus aureus (GBS) or Pseudomonas aeruginosa results in increased bacterial loads in the lungs and reduced macrophage phagocytic activity, highlighting SP-A1's important role in boosting macrophage phagocytosis and bacterial clearance. Additionally, SP-A1 helps inhibit bacterial infections by modulating inflammatory responses. It is also vital for pulmonary surfactant function, as SP-A1 binds to calcium ions to enhance the surface activity of lipid-extracted surfactants, thereby reducing alveolar surface tension and preventing alveolar collapse.
