Target Information
Nucleobindin-2 is a calcium-binding protein that may play a role in maintaining calcium homeostasis. It functions as a non-receptor guanine nucleotide exchange factor, binding to and activating the G-protein alpha subunit GNAI3. NUCB2 is integral to regulating energy metabolism through its peptide derivative, nesfatin-1. Nesfatin-1 is extensively expressed in both the central nervous system (like the hypothalamus) and peripheral tissues (including the pancreas, adipose tissue, liver, kidney, and intestine). Research indicates that nesfatin-1 influences food intake and energy expenditure by activating the melanocortin signaling pathway. Additionally, nesfatin-1 is involved in regulating insulin sensitivity. In a high-fat diet (HFD)-induced insulin resistance model, NUCB2 deficiency significantly worsened insulin resistance, underscoring NUCB2's crucial role in insulin sensitivity maintenance. Moreover, nesfatin-1 directly stimulates pancreatic β-cells to secrete insulin, demonstrating an insulinotropic effect. The expression of NUCB2 in immune cells, such as macrophages and T cells, is closely linked to metabolic inflammation. In HFD-fed mice, NUCB2 expression in macrophages and T cells within adipose tissue was notably increased, potentially associating with chronic low-grade inflammation and insulin resistance.
