Target Information
Dihydroorotate dehydrogenase (quinone), located in the mitochondria, catalyzes the conversion of dihydroorotate to orotate, using quinone as an electron acceptor. This enzyme is essential for UMP biosynthesis via the de novo pathway. In mice, DHODH plays roles in metabolic regulation, cancer therapy, immunomodulation, antiparasitic activity, and influences various physiological and pathological processes. DHODH inhibitors have been used in mice to explore their metabolic effects. For instance, in db/db mice, DHODH inhibitors enhanced glycemic control by reducing food intake and polydipsia, improving fasting blood glucose levels, and lowering non-fasting blood glucose and HbA1c levels.
