Target Information
Isoaspartyl peptidase/L-asparaginase exhibits both L-asparaginase and beta-aspartyl peptidase activity. It may contribute to the production of L-aspartate, which functions as an excitatory neurotransmitter in certain brain regions. This enzyme is particularly active with L-Asp beta-methyl ester. In ASRGL1 knockout mice, the thickness of the retina's outer nuclear layer (ONL) progressively diminishes, particularly after eight months of age, where this degeneration becomes more pronounced. Additionally, the inner segment (IS) and outer segment (OS) of the retina also thin out, and the rods and cones gradually degenerate. At eight months, ASRGL1 knockout mice display significantly weakened ERG responses, including reduced dark-adapted and light-adapted responses. These abnormalities are also evident in ASRGL1 mutant mice as early as three months of age, demonstrated by the loss of cone outer segments and decreased expression of rod and cone marker genes, such as Rhodopsin, Opn1sw, and Opn1mw.
