Target Information
Signal transducer and activator of transcription 1 (STAT1) is a signal transducer and transcription activator involved in mediating cellular responses to interferons (IFNs), cytokine KITLG/SCF, and various other cytokines and growth factors. Upon the binding of type I IFNs (IFN-alpha and IFN-beta) to cell surface receptors, signaling through protein kinases activates Jak kinases (TYK2 and JAK1), leading to the tyrosine phosphorylation of STAT1 and STAT2. STAT1 plays a crucial role in mouse skeletal development, particularly in bone formation and osteoblast differentiation. Research has demonstrated that STAT1 restricts osteoblast differentiation by interacting with the osteoblast transcription factor Runx2 and inhibiting its nuclear localization. Despite an increase in the number of bone-resorbing cells, a significant increase in bone density has been observed, indicating that STAT1 exerts an inhibitory effect on bone formation, which may surpass its role in inhibiting bone resorption.
