Target Information
Caspase-14 is a non-apoptotic caspase involved in epidermal differentiation, particularly in keratinocyte differentiation and is essential for cornification. It regulates epidermal maturation by proteolytically processing filaggrin. In mice, Caspase-14 primarily functions to oversee keratinocyte differentiation and skin barrier formation, while also shielding the skin from ultraviolet radiation damage. It plays a pivotal role in transforming keratinocytes into the stratum corneum, which is the skin's outermost layer composed of fully keratinized cells formed through programmed cell death, known as the cornification process. Research indicates that Caspase-14 processes profilaggrin into keratin, which subsequently promotes further hydrolysis of keratin into small peptides. These small peptides are crucial components of natural moisturizing factors, aiding in the maintenance of skin barrier function.
