Target Information

In cytotoxic T-cells and NK-cells, Granzyme B is a prevalent protease that induces caspase-independent pyroptosis when introduced into the target cell via the immunological synapse. In mice, Granzyme B facilitates the apoptosis of target cells by working in conjunction with perforin and plays a crucial role in immune defense and tumor immunity. Notably, mouse Granzyme B is 30 times less toxic than its human counterpart and can induce cell death without relying on the Bid protein. However, its toxicity can be restored by engineering its active site. Additionally, mouse Granzyme B exhibits greater specificity for certain substrates, such as pro-caspase-3.
