Target Information
Glutamate receptor ionotropic, NMDA 2B is a component of N-methyl-D-aspartate (NMDA) receptors (NMDARs), which serve as heterotetrameric, ligand-gated cation channels characterized by high calcium permeability and voltage-dependent blockage by Mg2+. The subunit of the NMDA receptor encoded by the GRIN2B gene plays a crucial role in the formation of cortical synapses both in early and late embryonic development. GluN2B expression is predominant during periods of rapid cortical synapse formation. In mice, the loss of GluN2B results in synaptic dysfunction, including defects in long-term depression (LTD) in the hippocampus, which is linked to cognitive impairments. In a mouse model of autism, mutations in GluN2B result in impaired synaptic function in the prefrontal cortex and hippocampus, leading to behavioral abnormalities and social deficits.
