Target Information

Zinc finger protein GLI1 functions as a transcriptional activator, binding to the DNA consensus sequence 5'-GACCACCCA-3'. It regulates the transcription of specific genes during normal development, playing a significant role in craniofacial and limb development, as well as in the development of the central nervous system and gastrointestinal tract. GLI1 is involved in mediating SHH signaling, influencing cell proliferation and differentiation through this pathway. In mice, GLI1's roles are diverse, encompassing tissue repair, inflammatory response, bone metabolism, tumorigenesis, and neural development. GLI1-deficient mice exhibit poor tendon-bone junction healing, reduced bone formation, and diminished biomechanical properties. In a rheumatoid arthritis (RA) model, GLI1 impacts macrophage polarization and the release of inflammatory factors by regulating DNA methyltransferases (DNMTs), thereby promoting joint inflammation and bone destruction.
