Beclin-1 is integral to the autophagy process, serving as a core subunit in various PI3K complex forms that facilitate the production of phosphatidylinositol 3-phosphate, thereby participating in multiple membrane trafficking pathways. Specifically, PI3KC3-C1 is involved in the initiation of autophagosomes, while PI3KC3-C2 plays a role in the maturation of autophagosomes and endocytosis. Beclin-1 is crucial in Alzheimer's disease (AD) models, where studies show that genetic reduction or loss of Becn1 function results in decreased neuronal autophagy, leading to neuronal degeneration, abnormal synaptic structures, increased amyloid β (Aβ) accumulation, heightened neuroinflammation, and cognitive dysfunction. Conversely, enhancing Becn1 expression or using Becn1 agonists significantly reduces Aβ accumulation, improves cognitive function, and extends the survival time of mice with AD.
