Target Information

Rat T-kininogen 1 is a 68 kDa acute-phase glycoprotein whose N-terminal cystatin-like domain inhibits cysteine proteases, while its C-terminal sequence releases T-kinin upon kallikrein cleavage, mediating rapid inflammatory vasodilation. A solvent-exposed 14-aa "KVE" loop (aa 218-231) between the two domains folds into a β-hairpin unique among rat kininogens, presenting a conformational epitope preserved after secretion and fixation. Affinity-purified antibodies targeting this loop trace native T-kininogen 1 in hepatic sinusoids, stimulated serum, and renal proximal tubules, delivering crisp cytoplasmic granules and single-band clarity on immunoblots with no cross-reaction to H-kininogen or α-2-thiol proteinase inhibitor.
