Target Information
Tacrolimus (FK506) is a potent immunosuppressant used to prevent organ transplant rejection. It is a macrolide lactone that binds to the FK506 binding protein (FKBP) to form a complex. Tacrolimus inhibits calcineurin, an enzyme involved in the production of interleukin-2 (IL-2). By binding to the immunophilin FKBP12, it forms a complex that inhibits calcineurin. This inhibition suppresses T-lymphocyte signalling and reduces IL-2 transcription.
