Target Information
Cyclin-dependent kinase inhibitor 1 plays a role in the p53/TP53-mediated inhibition of cellular proliferation in response to DNA damage. It binds to and inhibits cyclin-dependent kinase activity, preventing the phosphorylation of essential cyclin-dependent kinase substrates and halting cell cycle progression. It also aids in the nuclear localization and assembly of the cyclin D-CDK4 complex and enhances its kinase activity towards RB1. However, at higher stoichiometric ratios, it inhibits the kinase activity of the cyclin D-CDK4 complex. The absence of Cdkn1a affects the regenerative capabilities of mouse tissues. For instance, mice lacking Cdkn1a experience more pronounced functional impairment after cardiac injury, highlighting its role in cardiac repair. Additionally, Cdkn1a expression in skeletal muscle satellite cells is crucial for regulating muscle regeneration, and its absence leads to a reduced muscle regeneration capacity.
