Chicken Proto-Oncogene Tyrosine-Protein Kinase LCK (LCK) is a non-receptor tyrosine kinase that plays a crucial role in the activation and function of T lymphocytes. As a member of the Src family of protein tyrosine kinases, LCK is essential for the initiation of T-cell receptor (TCR) signaling, which is critical for T-cell development, activation, and immune response. LCK phosphorylates immunoreceptor tyrosine-based activation motifs (ITAMs) within the CD3 chains, leading to the recruitment and activation of downstream signaling molecules such as ZAP-70. This cascade ultimately results in the activation of transcription factors and the proliferation of T cells. The activity of LCK is tightly regulated by phosphorylation at key tyrosine residues, with CD45 and Csk playing critical roles in its activation and inactivation. Abnormal expression or activity of LCK has been implicated in various diseases, including cancer and autoimmune conditions.
