Mouse paraoxonase 2 (PON2) is a 43-kDa membrane-associated lactonase ubiquitously expressed in murine tissues, with enriched levels in liver, kidney, and vascular endothelium. It localizes to cell membranes and mitochondria, exerting antioxidant and detoxification activities by hydrolyzing lipid peroxides and xenobiotics, thereby protecting cells from oxidative stress. Critical for maintaining murine lipid homeostasis, vascular integrity, and mitochondrial function, it modulates inflammatory responses and cell survival pathways. Dysregulated expression associates with atherosclerosis, non-alcoholic fatty liver disease, and neurodegeneration in mouse models. As a key mediator of oxidative stress defense, mouse PON2 is a critical target for studying inflammatory and metabolic pathobiology, and developing therapeutics for cardiometabolic and neurodegenerative disorders.
