Target Information

Tissue factor (TF) initiates blood coagulation by forming a complex with circulating factor VII or VIIa, which then activates factors IX or X through specific limited proteolysis. TF is integral to normal hemostasis by starting the cell-surface assembly and propagation of the coagulation protease cascade. In mice, TF functions primarily in thrombosis, tumor angiogenesis, and inflammatory response. Beyond its role in coagulation, TF is involved in inflammation. For instance, in a sickle cell disease mouse model, excess heme triggers TF-dependent coagulation activation, leading to organ damage. The expression of TF in macrophages is closely linked to obesity-related chronic inflammation, indicating that TF might contribute to obesity-induced metabolic disorders by regulating macrophage function.
