Target Information
R-spondin-1 acts as a ligand for LGR4-6 receptors to activate the canonical Wnt signaling pathway. When it binds to LGR4-6 (LGR4, LGR5, or LGR6), these receptors interact with phosphorylated LRP6 and frizzled receptors, which are activated by extracellular Wnt receptors, thereby triggering the canonical Wnt signaling pathway and boosting the expression of target genes. RSPO1 is known as an intestinal epithelial stem cell growth factor that significantly enhances the Wnt/β-catenin signaling pathway's activity. Research indicates that RSPO1 activates the Wnt signaling pathway and fosters the proliferation and differentiation of intestinal stem cells by elevating the cell surface level of LRP6 (low-density lipoprotein receptor-related protein 6) and curbing DKK1/Kremen-mediated LRP6 internalization. Additionally, RSPO1 enhances the intestine's regenerative capacity following injury by promoting crypt stem cell proliferation and reducing apoptosis. In mouse models subjected to whole-body or abdominal radiation, RSPO1 significantly improved survival rates and repaired intestinal structure by bolstering the regenerative capacity of intestinal stem cells.
