Target Information

Tyrosine-protein kinase ITK/TSK is a tyrosine kinase crucial for regulating the adaptive immune response. It influences the development, function, and differentiation of both conventional T-cells and nonconventional NKT-cells. When antigen-presenting cells (APCs) activate the T-cell receptor (TCR), a cascade of phosphorylation events brings ITK to the cell membrane near the activated TCR, where LCK phosphorylates it. In mice, ITK (IL-2-induced tyrosine kinase) is primarily involved in TCR signaling, T cell differentiation, and immune response regulation. ITK is central to TCR signaling, enhancing T cell activation and proliferation by phosphorylating PLCγ-1, facilitating calcium influx, and activating MAPK. ITK-deficient mice exhibit impaired TCR signaling, leading to decreased IL-2 secretion, which in turn affects T cell proliferation and differentiation.
