Autoimmune regulator (AIRE) is a transcription factor crucial for promoting self-tolerance in the thymus by regulating the expression of various self-antigens known as tissue-restricted antigens (TRA), which are typically expressed in specific peripheral tissues. In mice, AIRE is vital for maintaining immune system balance and preventing autoimmune diseases. It helps protect against autoimmunity by regulating the expression of TSAs and facilitating the clearance of self-reactive T cells or their differentiation into regulatory T cells (Tregs). Mice deficient in AIRE exhibit abnormal TSA expression in the thymus, leading to the failure to effectively clear or suppress self-reactive T cells, thus triggering autoimmune responses against specific organs.
