Target Information

Beta-klotho plays a role in the transcriptional repression of cholesterol 7-alpha-hydroxylase (CYP7A1), the rate-limiting enzyme in bile acid synthesis. Although it is likely inactive as a glycosidase, it enhances the binding capacity of fibroblast growth factor receptors 1 and 4 (FGFR1 and FGFR4) to fibroblast growth factor 21 (FGF21). In mice, β-klotho has broad and complex functions, influencing metabolic regulation, anti-aging, anti-obesity, and kidney protection. A key role of β-klotho is in the regulation of bile acid synthesis. Research has shown that mice deficient in β-klotho exhibit significantly increased bile acid synthesis and excretion, accompanied by upregulation of genes involved in bile acid synthesis, such as Cyp7a1 and Cyp8b1. These findings highlight the critical role of β-klotho in maintaining bile acid homeostasis.
