Target Information
Dickkopf-related protein 4 (DKK4) antagonizes canonical Wnt signaling by inhibiting the interaction of LRP5/6 with Wnt and forming a ternary complex with the transmembrane protein KREMEN, which promotes the internalization of LRP5/6. DKK proteins are crucial in vertebrate development, where they locally inhibit Wnt-regulated processes such as antero-posterior axial patterning, limb development, somitogenesis, and eye formation. DKK4, as a Wnt signaling pathway inhibitor, impacts hair follicle development and differentiation by specifically inhibiting certain Wnt signaling pathways. In DKK4 transgenic mice, significant abnormalities in hair development are observed, including unusual hair morphology and disordered hair cycles. While DKK4 does not affect primary hair formation in wild-type mice, in Tabby mice, the DKK4 transgene completely obstructs the induction of secondary hair, leading to severe secondary hair deformities.
