Target Information
Cytochrome P450 7A1A is a cytochrome P450 monooxygenase that plays a pivotal role in the metabolism of endogenous cholesterol and its oxygenated derivatives, known as oxysterols. It operates by utilizing molecular oxygen to insert one oxygen atom into a substrate while reducing the other to form a water molecule, with two electrons provided by NADPH through cytochrome P450 reductase. In mice, the primary function of Cyp7a1 (cholesterol 7α-hydroxylase) is to convert cholesterol into primary bile acids, serving as the rate-limiting enzyme in the classical pathway of bile acid biosynthesis. Under conditions of a high-fat diet, the expression and activity of Cyp7a1 are suppressed, leading to cholesterol accumulation and liver inflammation. Conversely, mouse models that overexpress Cyp7a1 demonstrate resistance to obesity, insulin resistance, and fatty liver induced by a high-fat diet.
