Chicken Ephrin-A2 (EFNA2) is a cell surface GPI-bound ligand for Eph receptors, a family of receptor tyrosine kinases crucial for migration, repulsion, and adhesion during neuronal, vascular, and epithelial development. EFNA2 binds promiscuously to Eph receptors on adjacent cells, leading to contact-dependent bidirectional signaling into neighboring cells. This signaling is bidirectional, with forward signaling via Eph receptors and reverse signaling via ephrin ligands. EFNA2 is involved in various biological processes, including axonal orientation, cell adhesion and movement, synaptic development, and cell-matrix interactions. Dysregulation of EFNA2 has been linked to various diseases, including cancer, where it can promote tumor growth and metastasis.
