Target Information
Rat beclin 1 is a 60 kDa coiled-coil platform whose N-terminal BARA domain tethers the VPS34 lipid kinase complex to pre-autophagosomal membranes, nucleating isolation-membrane curvature. A solvent-exposed 18-residue "KVE" loop (aa 267-284) between helices 6 and 7 folds into a β-hairpin unique among rat BH3-only scaffold proteins, presenting a conformational epitope preserved after paraformaldehyde fixation. Affinity-purified antibodies targeting this loop trace native beclin 1 in hepatic autophagosomes, neuronal perikarya, and cardiomyocyte sarcoplasmic reticulum, delivering crisp punctate staining and single-band clarity on immunoblots with no cross-reaction to Bcl-2 or UVRAG.
