Target Information
Rat angiopoietin-3 (Angpt3) is a 58 kDa secreted glycoprotein whose N-terminal coiled-coil mediates oligomerization while the C-terminal fibrinogen-like domain binds Tie2 on endothelial cells, modulating vessel remodeling. A solvent-exposed 16-aa "KVE" loop (aa 345-360) within the fibrinogen domain folds into a β-hairpin unique among rat angiopoietins, presenting a conformational epitope preserved after glycosylation and fixation. Affinity-purified antibodies targeting this loop trace native Angpt3 in hepatic sinusoids, lung capillaries, and cardiac endocardium, delivering crisp extracellular deposition and single-band clarity on immunoblots with no cross-reaction to Angpt1 or Angpt2.
