Target Information

Mouse nucleobindin 2 (NUCB2) is a 53 kDa Ca²⁺-binding protein whose N-terminal DNA-like helix and two EF-hands form a Golgi-associated reservoir, cleaved to release nesfatin-1, an anorexigenic peptide. A solvent-exposed 15-aa "KVE" loop (aa 145-159) linking the second EF-hand and the leucine zipper folds into a β-hairpin unique among mouse nucleobindins, presenting a conformational epitope preserved after secretion and fixation. Affinity-purified antibodies targeting this loop trace native NUCB2 in hypothalamic neurons, pancreatic β-cells, and gastric endocrine cells, delivering crisp perinuclear and extracellular puncta with single-band clarity on immunoblots and no cross-reaction to NUCB1.
