Mouse cluster of differentiation 40 ligand (CD40L, also CD154) is a 39 kDa type-II trans-membrane glycoprotein that targets CD40 on B-cells, macrophages and dendritic cells. Binding induces trimerization, recruiting TRAF adaptors to activate NF-κB and MAPK cascades that drive immunoglobulin class-switching, germinal-center formation and dendritic-cell maturation. Its TNF-homology domain presents a unique, species-restricted trimeric epitope ideal for antibody generation; anti-CD40L immunoreagents detect membrane-bound and soluble forms in serum, lymphoid tissue and platelet-rich plasma without cross-reacting with other TNF-family ligands, providing a precise read-out of T-helper and platelet activation.
