Mouse mothers against decapentaplegic homolog 4 (SMAD4) is a 60 kDa common-mediator SMAD whose N-terminal MH1 domain clamps DNA while the C-terminal MH2 pocket captures phosphorylated R-SMADs, assembling transcriptional complexes. A solvent-exposed 15-residue "LPS" linker (aa 270-284) between MH1 and MH2 folds into a β-turn unique among SMAD isoforms, offering a fixation-stable epitope absent from SMAD2 or SMAD3. Affinity-purified antibodies targeting this linker trace native SMAD4 in nuclear speckles, cytoplasmic stress granules, and TGF-β-responsive epithelia, delivering crisp punctate staining and single-band clarity on immunoblots with no cross-reaction to other SMAD family members.
