Mouse high-mobility group nucleosome-binding domain 1 (HMGN1) is a 17 kDa nuclear protein whose nucleosome-binding domain clamps the 147 bp core, loosening linker histone contacts to enhance chromatin accessibility. A solvent-exposed 14-aa "KVE" loop (aa 40-53) between the two α-helices folds into a β-turn unique among mouse HMGN isoforms, presenting a conformational epitope preserved after fixation. Affinity-purified antibodies targeting this loop trace native HMGN1 in euchromatic foci, neuronal nuclei, and activated lymphocytes, delivering crisp punctate staining and single-band clarity on immunoblots with no cross-reaction to HMGN2 or HMGN3.
