Target Information

Mouse caspase-2 is a 50 kDa initiator caspase whose N-terminal CARD domain nucleates the PIDDosome, while the p18/p12 catalytic heterodimer cleaves Asp-containing substrates to trigger intrinsic apoptosis. A solvent-exposed 16-aa "KVE" loop (aa 347-362) between the large and small subunits folds into a β-hairpin unique among mouse caspases, presenting a conformational epitope preserved after activation and fixation. Affinity-purified antibodies targeting this loop trace native caspase-2 in oocyte cytoplasm, thymic cortex, and stressed fibroblasts, delivering crisp diffuse and punctate staining with single-band clarity on immunoblots and no cross-reaction to caspase-3 or caspase-9.
