Guinea pig ionized calcium-binding adapter molecule 1 (IBA1) is a cytoplasmic calcium-binding protein of the EF-hand protein superfamily, a specific marker for microglial cells. It features a conserved N-terminal EF-hand calcium-binding domain, central actin-binding motifs, and a C-terminal protein-interaction region critical for structural integrity and cytoskeletal rearrangement. It localizes to the cytoplasm of guinea pig microglia in central nervous system tissues, with negligible levels in neurons, astrocytes and non-neural cells. The protein mediates microglial motility and activation signaling, with conserved motifs for calcium coordination and intermolecular association. Guinea pig IBA1 shares core functional domains with mammalian orthologs, and has distinct species-specific sequence characteristics differentiating it from murine and rat IBA1.
