Target Information
Rat programmed cell death protein 1 (PD-1) is a 288-residue type I transmembrane glycoprotein belonging to the CD28 superfamily, featuring a single extracellular IgV-like domain, a 20-amino-acid stalk, and a cytoplasmic tail harboring two tyrosine-based signaling motifs. Expressed on activated T and B lymphocytes, myeloid cells, and select thymocyte subsets, the receptor undergoes phosphorylation upon ligand engagement, recruiting SHP-1 and SHP-2 phosphatases. Its expression is transiently up-regulated during immune activation and can be retained on exhausted lymphocytes. Antibodies targeting rat PD-1 specifically recognize the native extracellular epitope with minimal cross-reactivity to CD28, CTLA-4, or other IgV receptors, enabling precise detection in lymphoid tissues and cultured immune cells.
